Interpretive Summary:

Technical Abstract: Aluminum (Al) is one of the most important limiting factors for plant productivity on acid soils. We have begun characterizing major genes for Al tolerance in wheat and rye. We developed a genetic linkage map involving chromosome arm 4DL in wheat delimiting the AltBH gene to a 5.9 cM interval. In addition, AltBH was physically mapped to wheat chromosome arm deletion line 4DL breakpoints 0.70 and 0.86. The polymorphism rates for 4DL were extremely low and made further analyses difficult, therefore, map-based cloning of the homologous gene in rye chromosome 4RL (Alt3) was initiated. Using a rye F6 RIL population segregating for Al tolerance, five AFLP and two STS markers tightly linked to the gene were located. To construct a high-resolution map, the STS markers, flanking the gene at 0.4cM, were used to screen an 1123-line F2 rye mapping population. Markers spanning the Alt3 region covered a 10 kb region of a rice BAC, which contained four candidates genes, and one was expressed only in roots of rye under Al stress. A large-scale EST approach is also being analyzed for changes in gene expression in rye under Al stress. A total of 13 novel genes appeared to be regulated by Al-stress.
