Interpretive Summary: Technical Abstract: The W4 locus is one of the loci that control anthocyanin biosynthesis of soybean flowers and hypocotyls. The recessive mutable allele at the W4 locus, w4-m, was proposed to contain an autonomous transposable element. The w4-mutable line (w4-m w4-m) shows variegation in flower color. In a gene tagging study with the w4-mutable line, a number of mutants were found among progeny of the W4 wild-type revertants, such as female partial-sterile plants, malate dehydrogenase null, chlorophyll-deficient leaves, necrotic root, and male-sterile, female-sterile plants. By using SSR markers, the W4 locus was mapped to the end of molecular linkage group (MLG) D2 between Satt386 and the telomere with the distance between W4 and Satt386 as 2.7 cM. The eight mutants from the gene tagging study mapped all over the genome: female partial-sterile 1 (Fsp2), female partial-sterile 2 (Fsp3), female partial-sterile 3 (Fsp4), and female partial-sterile 4 (Fsp5) are located on MLG C2, A2, F and G, respectively; malate dehydrogenase 1 null and chlorophyll deficient (mdh1-n y20) are closely linked together on MLG H; male-sterile, female-sterile (st8) is positioned on MLG J; and another chlorophyll-deficient (CD-5) mapped to MLG E. |
