Interpretive Summary:

Technical Abstract: Insertion and expression of multiple antifungal genes has the potential of increasing resistance to a variety of fungal diseases. Using particle bombardment and bialaphos selection for the bar gene, multiple barley plants containing both a rice thaumatin-like protein (tlp) gene and a rice chitinase (chi) gene were regenerated from three transformation events. Southern analysis confirmed integration of the transgenes into the barley genome. Northern analysis of T0 plants indicated that event 1 did not contain RNA from either gene. Plants from event 2 expressed both genes at high levels, while plants from event 3 showed small amounts of tlp RNA but no chi RNA. Western analysis of T1 progeny confirmed that event 2 lines expressed both genes and event 3 lines expressed only tlp. Homozygous T2 lines have been identified and are being examined for gene expression levels for both transgenes.
