Interpretive Summary:

Technical Abstract: The soybean cyst nematode (SCN) Heterodera glycines is the most economically important pest of soybean worldwide, causing annual yield losses of $600 million. The developmentally arrested J2 stage comprises a vulnerable point in the nematode life cycle. Genes that control nematode development in response to environmental changes provide attractive targets for disruption. Hsp90 is thought to be an important and complex component of nematode stress response and developmental arrest pathways. The Hsp90 inhibitor geldanamycin (GA), produced by the bacterium Streptomyces hygroscopicus, was tested on heat shocked and non-heat shocked Caenorhabditis elegans eggs and on non-heat shocked eggs from three Heterodera glycines populations: NL1-RH, TN-17 and TN-18. We discovered that GA interferes with egg hatch and juvenile motility in both C. elegans and H. glycines. These results are the first demonstration of GA inhibition in any species of nematode, validating GA as a molecular probe of development and suggesting the potential of S. hygroscopicus as a novel biocontrol agent for nematodes.
