Technical Abstract: Oats are a rich source of polar lipids and oat polar lipids are being applied to some unique applications. However, many characteristics of polar oat lipid composition have not yet been characterized with modern methodology. Our objective was to identify constitutive lipids of the polar lipid fraction and estimate their relative abundance in the oat grain by HPLC-ELSD. We extracted total lipids from Morton (a low oil cultivar) and from Paul (a high oil cultivar) oats with a chloroform-methanol-water extraction. The crude oil was fractionated into neutral lipid, phospholipid and glycolipid fractions on silica SPE columns, and the quality of these separations was confirmed by carbohydrate and phosphorus analyses of these fractions. Major phospholipids in order of their relative abundance were phosphatidylinositol and phosphatidylcholine. The most abundant glycolipid was digalactosyldiglyceride. Polar lipid concentration was about the same in the high oil and low oil cultivars, the difference in total oil being comprised primarily of triglycerides. These results more clearly define the chemical composition of oat oil preparations being considered for value-added applications.
