Technical Abstract: A genomic library from Leptospira borgpetersenii serovar hardjo strain JB197 was prepared by mechanically shearing the DNA and inserting it into a positive selection vector. DNA was prepared from approximately 22,000 random clones and used as templates for automated sequencing. Sequence data was collected from either end of each insert and compiled into a series of contiguous sequences (contigs) using the program Phrap. These contigs were formatted into a searchable database that was used to search for the presence of selected genes using TBLASTN. We found genes homologous to those encoding several previously identified L. interrogans and L. kirshneri membrane proteins. Several Leptospira genes not described previously were also found by searching the database using partial peptide sequences obtained from membrane extracts of Leptospira interrogans serovar lai. By using the partial genomic sequence of Leptospira as a foundation to screen for genes encoding selected proteins we can rapidly gain insight into the genetic organization of each region. This study has resulted in identification of several genes that may be associated with virulence.
