Interpretive Summary: Technical Abstract: Microscopic analysis of Lygus hesperus alimentary canal revealed a short and narrow foregut, a midgut consisting of a 3 ventriculi, and a short hindgut which did not have epithelial cells. Two different cell types were observed in the midgut, columnar epithelial cells and granulated cells. The brush-border microvilli (BBMV) of the epithelial cells were notably longer in the region of the second ventriculus. Cry2AB did not demonstrate any biological activity in L. Hesperus feeding assays. However, association of Cry2Ab with the midgut was observed using immunocytochemical localization methods and competitive binding studies. In vitro incubations indicated that Cry2Ab associated with the midgut BBMV. Homologous competition binding assays performed with midgut homogenates indicated that the association was irreversible and specific (i.e. inhibited by an excess of the non-labeled form). |
