Interpretive Summary:

Technical Abstract: We initially screened several fungal cultures for xylanase production by solid substrate cultivation (SSC) on alfalfa. Aspergillus japonicus NRRL 360 produced 300 to 400 total IU/g substrate of xylanase, with small amounts of cellulase, and pectinase activity. On alfalfa, a moisture content of above 80%, particle size of 3 mm and harvest time of 9 days were erequired for SSC xylanase production from A. japonicus NRRL 360 at 27 to 3 deg C. Among various agricultural substrates tested, A. japonicus NRRL 360 exhibited high xylanase productivity on wheat bran, corn scover in addition to alfalfa. The xylanase extracts from NRRL 360 was active at pH 4.8 to 7.0, however, enzyme activity decreased significantly at pH 9.0.
