Technical Abstract: A high-resolution genetic map was constructed using the intermated B73 x Mo17 (IBM) mapping population. The population underwent random-mating for four generations resulting in a theoretical average three-fold increase in recombination events. One thousand low-copy-number RFLP markers and more than 500 simple sequence repeat (SSR) markers were mapped on the population. Low-copy-number RFLP markers from prior maps were used wheneve possible. In addition, a new PstI genomic library was constructed and 600 clones were sequenced and screened for copy number to identify additional SSR and low-copy-number RFLP markers. The SSR markers used included prior publicly-available markers and novel markers derived either from public sequence data or new enriched libraries developed in-house. Both prior- mapped RFLP and SSR markers will serve as a bridge between this map and prior public maps. In addition to the map itself, several other resources have been developed. They include 1) seed from the population will be available through the maize genetic stock center, 2) kits for PCR-based mapping on a subset of the population, and 3) access through the Maize Genome Database to the raw data used to generate this map. Current work is directed at anchoring this map to a maize physical map under development as part of the same grant.
