Technical Abstract: A factor(s) produced by the conceptus, which elicits a cascade of events during the process of maternal recognition of pregnancy, is required for the successful establishment of pregnancy. In ruminant animal species, this process is regulated through the expression of interferon-tau (IFN-tau) produced by the trophectoderm of peri-implantation conceptus. IFN-tau belongs to a multigene family similar to a group of Type I IFN genes. Among ovine IFN-tau (oIFN-tau) gene family so far characterized, individual oIFN-tau genes are not expressed to the same extent; up to 75% of oIFN-tau mRNA found in utero is based on the oIFN-tau o10 gene while others are weakly detected. To examine the regulatory mechanisms of oIFN- tau genes, the transcriptional activity of oIFN-tau genes was investigated using a transient transfection system and the upstream region of oIFN-tau genes that inserted into the chloramphenicol acetyltransferase (CAT) reporter plasmid (oIFN-tau-CAT). Experiments with serial 5'-deletions of the oIFNt promoter/enhancer region revealed that multiple enhancer elements existed between -654 and -555 bases of the oIFN?
