Citation: Interpretive Summary: The silverleaf whitefly is a major agricultural problem throughout tropical and subtropical regions of the world. Cells from the whitefly were inoculated with a virus (Chilo iridovirus) to determine if they could support virus growth. Observations made using light and electron microscopes indicated the virus could reproduce in the cells. Additional evidence indicated that virus proteins were being made in the whitefly cells.
Technical Abstract: A cell line from the silverleaf whitefly, Bemisia argentifolii Bellows and Perring (syn. B. tabaci, B biotype), was tested for its ability to support replication of the Chilo iridovirus. Several lines of evidence indicated that a productive infection was achieved. Infected cells became more rounded and developed cytoplasmic inclusions by day ten post infection. Virus particles were observed in defined areas of the cytoplasm using transmission electron microscopy. The expression of Chilo iridovirus structural proteins was detected using Western blot analysis and confirmed that a productive infection had taken place.
