One hundred ml of fragmented S. minor- mycelial suspension (1 g mycelia; fresh wt.) was added to each cup. Cups were placed in a humidity chamber at 100% RH for 5 days at 27 deg C. Cellophane bags were then carefully lifted, and outer surface of the cellophane facing the perlite was gently rinsed with cold running water for 2 min. Ten 1-cm**2 sections of the cellophane were placed on slides with the inner surface of the cellophane contacting the glass. Squares were stained with lactophenol cotton blue for 3 min, mounted in a glycerin gel, and infection cushions (IC) counted. IC/cm**2 of cellophane was 3.8 in response to Okrun, a sclerotinia-susceptible peanut cultivar, whereas the IC in response to Southwest Runner, a sclerotinia-resistant peanut, was 2.7 7with an LSD(0.05) of 0.5. |
