Interpretive Summary:

Technical Abstract: A quantitative trait locus (QTL) for meat tenderness in cattle has been previously described by Keele et al. To summarize, Warner- Bratzler shear force measurements were obtained at 2 and 14 days post-mortem on 294 progeny from a mating of a Brahman x Hereford bull to Bos taurus cows. The results of a genome scan identified a suggestive QTL on chromosome 15 with a peak at 28 cM from the most centromeric marker (MGTG13B). The 95% confidence interval for the QTL spans 23 cM on this chromosome from 17 to 40 cM. Efforts to fine map this QTL have focused on the discovery of microsatellite markers in the 23 cM interval and comparative mapping with HSA11. Markers that have been mapped to BTA15 have been used to screen YAC and BAC libraries for individual clones containing the marker. Individual clones were then subcloned and probed for microsatellites. In addition to subcloning YACs and BACs, bovine DNA was amplified in YACs using SINE-PCR. These fragments were then subcloned and probed for microsatellites. Human chromosome 11 shows conserved synteny with BTA15 and BTA29. Eleven coding genes from HSA11 were targeted as possible comparative candidate loci for the QTL or as markers to define breakpoints in conserved synteny between the two cattle chromosomes and HSA11. A Radiation Hybrid map has been constructed using the microsatellite markers that are included in the QTL interval. Coding genes were also typed on the Radiation Hybrid panel to test for synteny with BTA15 microsatellites and to develop a comparative map.
