Objective:
Carotenoids are indispensable to plants and play a critical role in human nutrition and health. The massive carotenoid accumulation in many fruits and vegetables depends on chromoplast biogenesis. However, the intriguing molecular mechanisms underlying chromoplast biogenesis remain an enigma. The objective for the cooperative research is to test whether chromoplast biogenesis is mediated by OR via regulating proteins in the plastid protein import pathway.
Approach:
Standard genetic, molecular and biochemical approaches will be employed. They include the examination of plastid import protein stability by western blot analysis, the selective import of chromoplast preproteins by import assays, the selective binding of chromoplast preproteins by selective binding assays, and the plastid membrane proliferation by microscope and biochemical analyses.
