Objective:
The objective of this collaborative project is the batch expression of multiple proteins of virus or insect vector origin. These proteins will be biologically characterized and used in diverse downstream assays.
The ultimate goal is to support development of diagnostic assay, experimental serology and countermeasure development. The assays developed will be very useful in investigating mechanisms of virus transmission and disease pathogenesis/virulence. Emerging vector-borne diseases are especially important because of their high frequency of emergence, their economic impact on the global livestock industries, and their zoonotic potential.
Approach:
ABADRU will provide sequences (and/or a construct in some cases) of the following proteins for preliminary expression and subsequent batch production in appropriate constructs:
Culicoides (Baculovirus expression)
D7 (~500 bp)
Late trypsin (~800 bp)
Maltase (~1800 bp)
BTV (E. coli expression)
VP-2’s of domestic serotypes 2, 10, 11, 13, 17 (~3 Kb)
for sero-diagnostics and antibody production
VP-7 of BTV-11 (~1200 bp)
for antibody production, cross-reactive for all BTV serotypes
EHDV (E. coli expression)
VP-2’s of domestic serotypes 1, 2, 6 (~3 Kb)
for sero-diagnostics and antibody production
VP-7 of EHDV-2 (~1200 bp)
for antibody production, cross-reactive for all EHDV serotypes
Other protein projects may be added as resources and finances allow.
