Objective:
To 1) perform transcriptome analysis of strawberry fruit ripening after various treatments; 2) conduct bioinformatic analysis of the transcriptome data to identify the common set of essential, and/or novel genes regulating phenolics accumulation; and 3) select two candidate genes for functional analysis.
Approach:
Several cDNA libraries will be constructed for RNA sequencing data. In silicon analysis and bioinformatic comparison of the expression profiles among different treatments will be carried out. The genes affecting phenylpropanoid pathways will be the focus on this study. From the candidate gene pool, two most promising genes will be cloned. The expression levels of these genes will be altered by overexpression and gene silencing techniques. The fruit quality and polyphenol content/profiles in the genetically modified fruits will be analyzed.
