Proposed studies will involve collection of field samples from suspect porcine epidemic diarrhea virus (PEDV) cases for use in developing in vitro methods to detect and propagate PEDV, the characterization of PEDV isolates for in vivo use, and animal studies. The project timeline will be 3 years; 30 months to conduct animal studies (which includes a 24-month appointment of an ORISE postdoc Fellow, veterinary), and 6 months to complete laboratory work, analyze data, and submit publications. A panel of PEDV field isolates and isolates from diagnostic laboratories will be prepared for genetic and possible antigenic analysis. PEDV infection models will include oronasal challenge of young pigs to investigate the pathogenesis of this disease. Study design will include estimating infectious dose as well as the kinetics of the infection. Young and adult swine challenge studies will be completed to study the onset and duration of the protective immune response which will include studies investigating passively acquired immunity in piglets. PEDV isolates found to have divergent antigenic profiles will be used to evaluate the extent of cross-protection following wild-type infection. In collaboration with the National Veterinary Services Laboratories, APHIS, tissue samples collected from experimental infections will be provided for evaluation of diagnostic methods.
