Within a segregating population the phenotype for tuber resistance will be determined by inoculating progeny of cross with a root+tuber resistant parent x root+tuber susceptible parent. A bulk segregant analysis using composited DNA from groups of resistant and susceptible parents will be conducted using simple sequence repeat (SSR), and single nucleotide polymorphism (SNP) markers. Furthermore, candidate genes extracted from analysis of the potato genome sequence will be screened. A bioinformatics pipeline will be developed to idnetify sequence reads that are unique to each pool. Candidate genes will be picked out sequenced potato genome to identify candidate genes and linked sequences using a Genome by Sequencing approach.
