Research Project: Northwest-Adapted Barleys for Resistance to Stem Rust (UG99)

Produce germplasm and potential varieties, with high end-use quality, that are resistant to the Ug99 stem rust pathogen and other diseases including Barley Yellow Dwarf Virus, scald, and stripe rust. Document and understand resistance and quality phenotypically and genetically; and make this germplasm available to interested cooperators.

Identify sources of resistance to Ug99 stem rust and other diseases. Cross these sources of resistance with adapted germplasm representing food, feed, and malt applications to develop GWAS and bi-parental mapping populations. Analyze and report results from the Cycle 5 GWAS population. Phenotype and genotype the Cycle 6 GWAS population Phenotype Cycle 6 for multiple phenotypes, including resistance to UG99, resistance to Barley Yellow Dwarf Virus (BYDV), stripe rust, scald, and other prevailing diseases, resistance to low temperature stress, vernalization sensitivity, grain physical parameters, and malting quality. Genotype the panel with genome-wide SNP markers, and with allele-specific markers. Integrate the genotype and phenotype data via genome wide association scanning methods. Develop new germplasm (Cycle 7) using doubled haploid and speed breeding techniques. Build on our full genome sequencing of 8 genotypes and focusing on 5H (where target genes/QTLs for resistance, abiotic stress resistance, and quality are located) to include long-read sequencing of one genotype. The germplasm and data will be available to interested cooperators for further testing, analysis, gene discovery, and cooperative release for commercial production. This NACA supports research conducted under National Program (NP) Number 303, Plant Diseases. The research is conducted as part of ARS research project 2072-22000-042-00D, funded by Program Increases FY10, FY17, FY19, and FY20 for Small Grain Genomics (Barley Double Haploid).
