Objective:
The objectives of this project are: 1) Map genes controlling cold acclimation and deacclimation in canola; and 2) Determine the role, if any, for candidate genes identified by GWAS using arabidopsis as a model.
Approach:
1. Map genes controlling cold acclimation and deacclimation in canola. We will cross canola line with high and low deacclimation rates along and phenotype the resulting progeny using our previously established protocols. Additionally we will cross a line with superior freezing tolerance and slow deacclimation rate to several advanced spring breeding lines. The resulting progeny will also be phenotyped. Additional seeds from each confirmed cross will be allowed to self pollinate to generate an F2 population. These F2 populations will be saved for further study.
2. Determine the role, if any, for candidate genes identified by GWAS using arabidopsis as a model. We have obtained lines of arabidopsis containing mutations in orthologous genes to those we suspect of playing a role in deacclimation processes in canola. We will phenotype these mutant arabidopsis lines using previously established protocols to determine if the mutations impact freezing tolerance and deacclimation rates in arabidopsis.
