Objective:
1. Identify DNA markers associated with rust resistance genes in wheat, 2. Identify wheat genes that mediate susceptibility to wheat leaf rust, and 3. Identify leaf rust pathogen avirulence gene products that are recognized by wheat resistance genes.

Approach:
The exome capture method will be used to identify genome-wide DNA polymorphisms in a representative set of wheat experimental lines and cultivars. These data will be used to develop a haplotype-based method of postulating the presence of rust resistance genes in new varietal candidate wheat lines. Wheat susceptibility genes for leaf rust will be identified and tested using a variety of functional genomics approaches including RNA-Seq, EMS mutant panels, and gene silencing experiments. Rust avirulence gene products that trigger resistance reactions in the plant will be identified using comparative genomics, EMS mutants, and functional assays.
