Research Project: Screening Xylella fastidiosa in the USDA ARS National Collection of Genetic Resources for Carya

Objective:
1) Refine diagnostic procedures to insure identification of infection; 2) Refine sample collection protocols to optimize both phenotypic recognition and diagnostic identification; and 3) Screen collections of the NCGR-Carya to identify infected and disease free trees.

Approach:
1) Isolates of Xylella fastidiosa from previous work in pecan at LSU, Shreveport and TAMU, Uvalde will be sequenced; 2) New PCR primer sequences will be designed from the pecan Xf sequence and tested against the pecan genome to ensure that they do not bind non-specifically; 3) Detailed PCR conditions will be defined, specifying polymerase to be used; and 4) LAMP technique to identify Xf in other plant species will be evaluated using pecan DNA and pecan sap.
