To 1) develop comprehensive datasets on the levels of primary and secondary metabolites in maize grain and tomato fruits; 2) identify and quantify minor metabolites.
Untargeted Nuclear Magnetic Resonance (NMR) based metabolomics approach including unbiased data processing will be applied to generate a comprehensive view of the relative levels of primary and secondary metabolites in grain crops and fruits. Two-dimensional NMR spectra will be acquired and processed. The spectral data will be compared with literature data and on-line databases to assign the signals of unassigned compounds and, if necessary, the corresponding standards will be added to confirm the assignment. Relative or absolute quantification of metabolites will be carried out and the data will be subjected to a proper multivariate statistical analysis. In this way, valuable information on the overall metabolome differences and similarities between different genotypes, and influence of cultivation practices will be assessed.
