Objective:
The objective of the project is to determine the collateral variation in seed composition between crop plants developed using genetic engineering, mutagenesis, and classical breeding. The goal is to compare and characterize protein profiles of a group of 30-50 soybean (Glycine max) genotypes that will include cultivars as well as a diverse group of G. max and G. soja germplasm accessions, fast neutron (FN) mutants, and transgenic soybeans.

Approach:
Several experimental approaches will be used to identify seed proteins. Seed proteins will be extracted, and further separated using 2-dimensional polyacrylamide gel electrophoresis (2D-PAGE). Separated proteins will then be visualized by staining and subsequently scanned and further evaluated using protein expression software. Differences in protein profiles will be quantified with an image analyzer and peptide fragments will be determined using matrix-assisted laser desorption/ionization-time of flight mass spectrophotometry (MALDI-TOF-MS). The proteins will be identified using the NCBI non-redundant and SWISS-PROT databases.
