The overall objective is to understand iron acquisition from cattle during infections causing Johne’s disease. Accomplishing this objective will increase our understanding of the pathogenesis of this pathogen and perhaps identify intervention points to prevent infection of dairy cows, sheep and goats.
Studies will investigate multiple mechanisms by which MAP regulates environmental and intracellular iron, and thus understanding these mechanisms will strengthen our ability to control Johne’s disease. Succinctly put, we will perform experiments to define the iron-induced transcriptome and proteome of MAP. Methods used in this approach include: iTRAQ analysis, RT-PCR, DNA microarray hybridization, DNA footprinting analysis, recombinant protein expression and allelic exchange to create a gene knockout mutation.
