The goal of this project is characterizing the plant defense responses of tepary bean genotypes to BCMV infection by RNAseq analysis to recover host and viral profiles associated with gene expression during the infection cycle and signatures indicative of post-transicptional and transcriptional gene silencing. This proposal focuses on genome characterization of selected tepary bean associated Bean Common Mosaic Virus (BCMV) isolates, and characterization of tepary bean host-BCMV interactions by dual host pathogen sequencing.
Obj. 1 Genome characterization of selected tepary-bean associated BCMV isolates. The complete genome will be determined for eleven BCMV isolates originating from tepary bean accessions and/or land races, and three Arizona land races. The genomes will be (a) characterized by analyzing genomic variability at the nucleotide and protein levels, (b) analyzed phylogenetically to understand the relationship(s) with other well-characterized BCMV isolates, and in the context of potential co-evolution with tepary bean vs. common bean. Viral genome sequences will be aligned at the nucleotide and amino acid levels to identify differences and similarities among the isolates, including at the SNP level. Relationships will be investigated by phylogenetic analysis by alignment with BCMV reference sequences from different locations, worldwide, available in the GenBank database.
