Objective:
Design and develop an enzyme linked-immunosorbent assay (ELISA) to efficiently and sensitively detect antibodies to the foreign epitope named S-Tag in vaccinated swine when the S-Tag is part of a modified porcine reproductive and respiratory syndrome virus (PRRSV).

Approach:
An ELISA that was previously developed by the Cooperator for the S-Tag will be used to determine if the ELISA will work using swine serum from animals inoculated with a PRRSV modified live virus S-Tag vaccine (MLV-S-Tag) developed by ARS. Swine serum has been shown to be technically difficult to work with in many other ELISA assays, due to inherent high background. If this ELISA approach does not work, Cooperator will develop a different ELISA (sandwich ELISA) using one or more anti S-Tag monoclonal antibodies previously developed.
