Research Project: Elucidating Factors Impacting Skin Set and Periderm Development via Metabolomics and Genomic Analysis

Objective:
To investigate the molecular and biochemical mechanisms impacting potato periderm development and post-harvest storage quality within a potato population segregating in skin set.

Approach:
For phenotyping and association mapping, 200 to 300 progeny from a biparental cross of parental genotypes contrasting for skin set will be generated in the greenhouse and grown in multi-hill plots under MN field conditions. Foliar tissue samples will be submitted for SNP (single nucleotide polymorphism) genotyping using the V3 Illumina Array, and association analysis will be performed using ClusterCall, rrBLUP and GWASpoly. At harvest, genotypes will be assessed for maturity, skin color, skin set. Tubers will be provided for metabolite profiling at harvest and through postharvest suberization. Monoterpenoid volatiles, polar amino acids, or hydrophobic lipids profiles will be assess using LC/MS untargeted metabolic profiling; UHPLC-ESI-QTOF/MS.
