Objective:
The objective of this proposal is to develop a recombinant pig via site-directed genetic modifications that are resistant to zoonotic influenza A viruses (IAV) and which will not transmit swine influenza.

Approach:
Genome editing tools and innovative influenza resistance tools will be employed to engineer influenza resistance in pigs. Aim-1 will assemble and test composite vectors for targeting IAV replication, assembly, and dissemination of swine adapted IAV strains ex vivo. Aim-2 will create genome edited pigs designed to be resistant to IAV infection and transmission. Aim-3 will challenge the recombinant pigs with swine adapted IAV subtypes and assess viral production and transmission rates.
