The overall objective of this research project is to develop a genome editing tool for trait modification in grape cultivars. This objective will be accomplished in phases. During 2019-2020, the key objective is to propose new approaches for improving the direct delivery methods.
Experimental procedures for accomplishing the objective involve many steps, including design and construction of appropriate CRISPR-Cas9 systems, preparation of appropriate target embryogenic callus and delivery of the CRISPR-Cas9 systems into the callus cells,and evaluation of the resulting callus cells and/or vines for confirmation of the success of gene editing. We will attempt to edit the V. vinifera VvMybA1 gene in this proposed project. We will choose the wine grape V. vinifera cv. Chardonnay and the table grape V. vinifera cv. Thompson Seedless as the initial target germplasm.
