Research Project: Potential Role of Dietary Mushroom in Promoting Brain Health

Location: Diet, Genomics and Immunology Laboratory

Project Number: 8040-51530-058-007-R
Project Type: Reimbursable Cooperative Agreement

Start Date: Jan 7, 2020
End Date: Jun 30, 2021

Objective:
To identify the changes in pig brain gene expression with dietary supplementation with white button mushroom using RNA sequencing. To identify metabolic pathways related to Carbohydrate and amino acid metabolism as well as Reactive oxygen species (ROS) and inflammatory response in brain tissue after dietary supplementation.

Approach:
Archived brain samples from Hippocampus, Hypothalamus and Cerebral Cortex derived from pigs fed Mushroom-supplemented diets will be used for whole transcriptome analysis following isolation of RNA and sequencing. Mapped reads for each sample will be summarized into gene expression levels and treatment groups will be compared against non-dietary intervention group. Global metabolomic profiling of Carbohydrate, aminoacid, nucleotide and energy metabolim will be performed in brain samples using Gas Chromatpgraphy(GC)-Mass Spectrometry(MS). Transcriptomic and metabolomic data will be integrated with previously generated microbiota abundance data using Pearson correlation analysis.
