Objective:
Cooperator will evaluate the effects of sorghum extracted compounds on metabolic cancer processes using tissue culture models.

Approach:
Sorghum bioactive compounds will be extracted and isolated at Grain Quality and Structure Research Unit from various sorghum genotypes and sent to Towson University. Cooperator will evaluate the effects of these extracts and isolated compounds will be evaluated on cancer cell models to study the effects on cancer cell metabolism. Polyphenols will be extracted from two types of sorghum bran by the ARS. Human colon cancer cells will be incubated with sorghum extract (1.25 mg extract/mL medium) for up to 48 hours. RNA will be extracted and reverse-transcribed. mRNA expression of carcinogen-activating and -inactivating enzymes, enzymes involved in epigenetic regulation, and enzymes involved in the cellular antioxidant response will be quantitated as mRNA using qPCR and/or next generation sequencing, as protein using western blotting, and protein activity will be measured with catalytic activity assays if possible. Epigenetic changes will also be looked at using specific kits and/or next generation sequencing.
