Objective:
The principle objectives of the project are: 1) Evaluate (index) seed inventories in the USDA/ARS Capsicum germplasm collection for the presence of Potato Spindle Tuber Viroid (PSTVd), 2) Sequence the PCR product(s) of samples that test positive and compare with known strains of PSTVd, and 3) Update the GRIN-Global database to reflect the test results for all indexed seed inventories.

Approach:
Approximately 1,000 seed inventories (200 seed each) will be drawn from the Capsicum germplasm collection (Griffin, Georgia), packaged, and sent to AgDia Testing Services. All samples will be indexed for the presence of Potato Spindle Tuber Viroid (PSTVd and related Posiviroids) using AgDia's proprietary Pospiviroid [Polymerase Chain Reaction (PCR-based)] test. The PCR products of all samples testing positive will be sequenced to identify specific Pospiviroids (PSTVd and others). All test results and sequence data will be returned to the Griffin, Georgia, site and used to update the GRIN-Global database.
