We plan to test whether populating the mammary gland with an enriched population of mesenchymal stromal cells (MSCs) will promote the healing of existing infections and increase resistance to new infections in dairy animals with mastitis.

Prior to the initiation of a large scale clinical trial in cows, we will examine the potential therapeutic effect of MSCs in tissue culture and mouse models. Aim 1: Isolation, expansion and characterization of mouse and bovine MSCs Aim 2: Functional characterization of the immunomodulatory effect of MSCs on mammary epithelial cells in tissue culture conditions Aim 3: Evaluation of the modulation of mammary inflammation by MSCs
