1) Characterize the protein content and arrangement of the toxin biosynthetic apparatus in the Fusarium head blight pathogen Fusarium graminearum; 2) Test for differentiation of the endoplasmic reticulum for specific primary and secondary terpenoid metabolite pathways upon toxin induction.
Our approach will be to identify components of the toxin biosynthetic apparatus by tagging proteins with fluorescent proteins and then localization using confocal- or superresolution microscopy. Protein-protein interactions will be accessed using fluorescence resonance energy transfer utilizing acceptor photobleaching. Site directed mutagenesis will allow for measuring the effect of individual proteins on toxin accumulation.
