Research Project: Phenotyping and Genotyping of RIL Populations for Gene Discovery and Marker Development

Objective:
1. Pyramid early and late leaft spot resistance in elite cultivars using resistant recombinant inbred lines (RILs) identified in the Florida-07 x GP-NC WS 16 population as the trait donors. Molecular markers associated with resistance to early and late leaf spot resistance will be used to facilitate these breeding efforts. 2. Florida-07 x C76-16 population was genotyped on version 2 array. Genetic map will be constructed. Additional phenotyping for drought tolerance and aflatoxin resistance will be performed this year. 3. Tifrunner x C76-16 population was genotyped on version 2 array. Genetic map will be constructed, additional phenotyping for drought tolerance will be performed this year. 4. Genetic mapping of white mold resistance in Tifrunner x NC 3033 will be performed. Mapping results will be compared with that of QTL-seq analysis. Field trial to confirm the Quantitative Trait Loci (QTLs) for white mold resistance will be carried out this year. The Isleib half of this population will also be phenotyped for Rhizoctonia limb rot, a disease that has reemerged in recent years. An additional year of field resistance to tomato spotted wilt virus (TSWV) will also be collected. 5. Initial phenotyping of the Florida-07 x Florunner population for resistance to TSWV, yield and grade. Genetic and phenotypic data will be analyzed to determine if molecular markers previously identified in other populations will also be effective in this population. 6. A second year of phenotyping of the Tifrunner x OLin population (162 lines) for resistance to Sclerotinia minor will be conducted. Genetic mapping of Sclerotinia resistance in Tifrunner x OLin will be performed. 7. Customization and loading of a database and web interfaces for genetic map, marker, phenotyping, and QTL data are in progress. Enriched genetic information will be added to the Peanut Breeder's Toolbox. 8. Intertek platform has already been established. Single Nucleotide Polymorphism (SNP) markers associated with multiple traits were recruited. Ten most informative SNPs markers that can be used for multiple breeding programs will be prioritized for service.

Approach:
Recombinant inbred line (RIL) populations will be genotyped and phenotyped. Data will be analyzed to attempt to develop molecular markers for resistance to white mold, tomato spotted wilt virus, and drought. Previously identified molecular markers will be used to facilitate breeding for resistance.
