Research Project: Extension and Research to Combat Insecticide-Resistant Soybean Aphids

Location: Corn Insects and Crop Genetics Research

Project Number: 5030-22000-019-002-N
Project Type: Non-Funded Cooperative Agreement

Start Date: Oct 1, 2017
End Date: Sep 30, 2020

Objective:
This study aims to define the level and geographic range of pyrethroid resistance in Iowa. Furthermore, the biochemical and genetic basis of pyrethroid resistance will be investigated.

Approach:
Field populations will be collected from several locations across NW Iowa, and then screened in the laboratory for running diagnostic bioassays that will estimate the frequency and fold-resistance compared to a susceptible baseline colony. The genomic basis of pyrethroid resistance will be investigated by the application of next generation sequencing (NGS) technologies by re-sequence pools of resistant and susceptible individuals from two different locations. Mutations that are fixed or showing a high frequency differences between resistant and susceptible pools will be identified computationally, and genes or genome regions containing these mutations will be identified for further downstream research.
