To 1) develop comprehensive datasets on the levels of phytohormones in tomato fruits; 2) identify and quantify their metabolic forms.
Phytohormones in plant tissues will be analysed by HPLC-ESI-MS/MS developed and routinely used in the Co-PI laboratory. Briefly, after extraction, purification and addition of appropriate internal standards, quantification will be done on HPLC (Ultimate 3000, Dionex, USA) coupled to hybrid triple quadrupole/linear ion trap mass spectrometer (3200 Q TRAP, Applied Biosystems, USA) set in selected reaction monitoring mode by using a multilevel calibration graph with stable isotope labelled internal standards. This method allows simultaneous, reliable and precise identification and quantification of more than fifty metabolic forms of aforementioned phytohormones in one single sample (ca. 50 to 100 mg FW needed for one biological replicate). In this way, valuable information on the overall phytohormone differences and similarities between different genotypes, and influence of cultivation practices will be assessed.
