Research Project: Assessing Genetic Diversity in the Cultivated Strawberry (Fragaria ×ananassa) Collection at the National Clonal Germplasm Repository

Objective:
1) Genotype up to 333 strawberry (Fragaria x ananassa) accessions from the National Clonal Germplasm Repository Collection with the strawberry single nucleotide polymorphism (SNP) array. a) Assess the diversity in this collection. b) Redefine a core set of accessions using molecular analysis. 2) Confirm the pedigree records of cultivars using molecular data from up these SNPs. 3) Establish a detailed “genetic fingerprint” record for each accession.

Approach:
DNA will be extracted for the F. ×ananassa accessions in the summer of 2017. They will be genotyped and analyzed in combination with genotypic data from accessions from the NCGR. The genotypic and phenotypic data will be entered into the Germplasm Resources Information Network (GRIN). Genetic diversity will be assessed using the R with the phylogenetics package Poppr and diversity indices will be calculated using the population biology package VEGAN. Parentage will be confirmed and inferred where possible using the parentage assignment software CERVUS and a Microsoft Excel-based tool. Accessions representing the allelic diversity in the collection will be selected based on the advanced M strategy using a modified heuristic algorithm implemented in PowerCore software. The 160 F. ×ananassa accessions that were not previously evaluated by the UC breeding program will be propagated and shipped to the UC Davis to phenotype.
