1) Backcross new sources of resistance to all three rust species into elite hard winter wheat lines to create new germplasm lines or cultivars with durable, broad-spectrum rust resistance. 2) Develop populations to map and/or evaluate rust resistance genes. 3) Test resulting lines and/or populations for rust resistance and agronomic performance.
One to several resistance genes at a time will be backcrossed using marker-assisted selection into elite, adapted hard winter wheat lines. Recombinant inbred populations, doubled haploid populations, or near isogenic lines will be developed to map and evaluate rust resistance genes. Backcrossing, near isogenic line development, and marker development will be combined when possible. High throughput genotyping will use either KASP markers or genotyping-by-sequencing (GBS). The resulting lines or populations will be phenotyped for rust resistance reactions to all three rusts in the field and/or greenhouse. The effect of resistance gene alleles or gene combinations on phenology, yield, test weight, and grain quality will be determined.
