Objective:
This project is a continuation of a previously funded project whose objectives are to develop RT-PCR and real-time RT-PCR assays for exotic viroids, including Coconut cadang-cadant viroid (CCCVd) and tomato apical stunt viroid, among others, and conduct confirmatory RT-PCR detection tests to provide species level identification for each sample. Species-specific RT-PCR primers will also be designed and evaluated to examine their specificity and applicability as a diagnostic method for the CCCVd and TASVd in the United States, so that introduced viroids can be efficiently detected and distinguished. The Cooperator will coordinate the agreement and sampling and shipment of samples to ARS.

Approach:
ARS will select the best methods to use for palm leaf storage and for rapid and reliable extraction protocols for total nucleic acids from palm leaves. ARS will develop RT-PCR and real-time RT-PCR assays for CCCVd and TASVd and conduct confirmatory RT-PCR detection tests to provide species level identification for each sample. Species-specific RT-PCR primers will also be designed and evaluated to examine their specificity and applicability as a diagnostic method for the CCCVd and TASVd in the United States, so that introduced viroids can be efficiently detected and distinguished. ARS will test palm samples sent from US territories for the presence of CCCVd and related viroids.
