To validate that the affinity column method can accurately determine the naturally occurring individual folate vitamers and quantify them in raw peanut seeds.
In plants, the vitamin folate occurs in many different forms and complexes with glutamates. The human body is not capable of using all forms so it is necessary to determine which forms are present in peanuts to evaluate the quality of peanuts as a source of naturally occurring folate. Raw peanut seeds will be digested with a variety of enzymes to free the folates from their binding sites. Affinity chromatography will be used to trap the folates so that other materials may be removed from the digests. The folates will then be eluted from the column and identified and quantified by high performance liquid chromatography. Results will be compared with analysis using the traditional microbiological method which does not distinguish between folate forms. Results will be used to determine if this methodology can be applied to large numbers of samples for screening of current commercial peanut varieties and the germplasm collection.
