Determine the genetic diversity of aflatoxigenic isolates of Aspergillus flavus and Aspergillus parasiticus in order to use that information for the design of RNA interference technology to reduce aflatoxin in peanuts and corn.
Samples of soil and contaminated peanut and maize from Uganda and Kenya will be processed to isolate Aspergillus spp at the laboratory in Lilongwe, Malawi. The fungal isolates will be identified and genomic DNA will be extracted and sent to the National Peanut Research Laboratory (NPRL) in Dawson, Georgia, USA, for sequencing. The NPRL will arrange for the high throughput sequencing of the samples, generate a database to be hosted at the NPRL, and use the information to design molecular constructs for RNA interference to silence aflatoxin synthesis.
