Adaptive immune response to porcine reproductive and respiratory syndrome virus (PRRSV) infection is characterized as weak and delayed, but the mechanism by which this happens has not been determined. Overall working hypothesis is that PRRSV infection causes a depletion of circulating lymphocytes (data already obtained) and effect normal thymic physiology such that egress of naive T cells from the thymus decrease. The purpose of this work is to evaluate the effects of infection on thymic gene expression and evaluate permissivity of thymic cells to PRRSV infection. The results will provide additional data for hypothesis and further studies on the mechanism in which PRRSV attenuates host adaptive immunity.
Map gene expression changes in thymus collected from pigs 4 or 10 days following infection with different porcine reproductive and respiratory syndrome virus (PRRSV) strains and evaluate histologic changes associated with infection. Correlate results to disease severity and impact on host adaptive immunity. Determine if a particular cell in the thymus can support PRRSV replication and evaluate changes in gene expression following exposure of thymic cells to PRRSV in vitro.
