1. Develop C. hystrix introgression lines aiming to transfer of downy mildew and root knot nematode resistance into cultivated cucumber. 2. Understand the mechanisms of homologous recombination between cucumber and C. hystrix chromosomes.
1. Development of C. hystrix introgression lines (ILs). 1.1. The C.hystrix genome will be sequenced; molecular markers (SSRs, Indels) will be developed to construct a linkage map for C. hystrix; 1.2. ILs will be developed through backcrossing of amphidiploid between cucumber and C. hystrix with cucumber; 1.3. ILs will be characterized with molecular markers and cytological tools (fluorescence in situ hybridization) and chromosome analysis. 1.4. ILs will be screened for downy mildew and southern root knot nematode resistance. 2. Understand mechanisms of homoeologous recombination. Chromosome pairing in various stages of meiosis in the interspecific F1 hybrid between cucumber and C. hystrix (haploid) will be investigated to clarify underlying mechanisms of recombination between homoeologous chromosomes. 3. Graduate student training One graduate student and one postdoc will be trained.
