To perform a targeted analysis of gene expression of the epithelial cells lining the sperm-storage tubules found in the chicken oviduct to better understand the micro-environment required by sperm for prolonged storage. This information will then be applied to the formulation of novel media to store poultry semen in vitro.
Sperm storage tubules (SSTs) will be isolated from chicken hens of known high and low fertility and from virgin hens. For the targeted analysis, the cells lining the SSTs from each group of chickens will be isolated using laser capture microdissection technology. RNA will be extracted from the isolated SST cells, amplified, and then sequenced with Illumina Next Generation deep sequencing. SST transcripts will be identified and genes of interest, such as those associated with lipid metabolism and antioxidant activities, identified.
