The two objectives of the proposed research are: (1) to identify molecular markers that can be used to assess population structure of Uromyces transversalis and (2) to develop diagnostic assays for U. transversalis detection.

Whole genome sequencing will be used to identify molecular markers that can be used for population studies and molecular diagnostic development. Once identified, molecular markers will be tested on a collection of U. transversalis isolates. Genetic diversity will be assessed, and if possible, used to determine the origins of isolates collected in California and Florida. Markers suitable for diagnostic development will also be tested for the rapid and accurate detection of the pathogen. Preparation of U. transversalis isolates and isolation of high-quality genomic DNA will be performed at the USDA/ARS/FDWSRU facility, at Ft. Detrick, MD, in the necessary containment laboratory/greenhouse facilities, and the sequencing will be performed by collaborators at the University of Georgia. Post-sequencing analysis will be performed jointly between USDA/ARS and the University of Georgia. Testing of diagnostic assays will be performed by ARS staff at Ft. Detrick.
