Optimization of protein extraction from manure by a solvent extraction method. The extracted proteins could be used by Mitsubishi Chemical for production of ammonia, biofuels, and feed additives (amino-acids) for excess N mitigation process.

The research will optimize protein extraction from manure by the solvent extraction method in terms of pH by using NaOH as the solvent. The protocol for the solvent extraction will be provided by The KAITEKI Institute, Inc. (TKI). The research project will be conducted on anaerobic digestion effluent from dairy operation. The sample will be also provided by TKI. Protein extraction of manure will be determined in multi-step chemical fractionation of the manure. Condition parameters to optimize include concentrations and pH in a range from 3 to 11. The protein content (Cp) will be determined from the total nitrogen and NH4+ concentrations through the Kjeldahl method. Protein extraction efficiency (EE) will be determined as the percent ratio of the sum of Cpi and Cp. The research project will be conducted on anaerobic digestion effluents from dairy operation. The approach will be done using bench scale experiments at ARS-Florence. Initial manure samples containing protein feedstock will be characterized for metals (ICP), chemical oxygen demand, alkalinity, electrical conductivity, and nutrient analyses (NPK).
