Determine the stability of zeaxanthin content in orange-fleshed diploid potatoes grown in two different environments (Maine and Florida). Determine the genetic composition of 70 4x-2x potato hybrids for CHY2.3 and Zep1 alleles. Develop tetraploid orange-flesh (high zeaxanthin) and intense yellow-flesh (high lutein) potato germplasm.

Eighteen orange-flesh, high zeaxanthin diploid (2x) clones from Solanum phureja-S. stenotomum plus the check varieties Yukon Gold and Peter Wilcox will be grown in Presque Isle, ME and Hastings, FL in 2013 in a randomized complete block design with three replications of four hills per clone. Weather parameters will be monitored during the growing season in both locations. Tubers will be harvested and shipped to Beltsville, MD. Carotenoids will be extracted within one month of harvest and analyzed by high performance liquid chromatography. Approximately 70 4x-2x hybrids between yellow-flesh tetraploids and orange-flesh diploids will be screened for the presence of the CHY2.3 and Zep1 alleles using allele-specific markers. All of the 4x-2x hybrids are expected to have at least two copies of Zep1. Not all of the 4x-2x hybrids may have CHY2.3. 4x-2x hybrids possessing both CHY2.3 and Zep1 will be intercrossed to generate a population segregating for yellow-and orange-flesh. These hybrids will also be crossed with other diploid orange-flesh clones.
