Screen potential new varieties for Potato virus Y (PVY) resistance using molecular markers in West, Midwest, and Eastern potato breeding programs in the U.S. Publish a list of resistant parents and their associated resistance genes.

Markers currently available and associated with extreme PVY resistance are the SCAR marker RYSC3 for the Ryadg gene and the STS marker YES3 for the Rysto gene. These markers and other appropriate markers for these genes will be used to screen a set of parent varieties currently used and other varieties that have reported PVY resistance. This screening will be done by a team of scientists at the USDA, Agricultural Research Service (ARS), Aberdeen, Idaho; University of Maine, Orono, Maine; and, Michigan State University, East Lansing, Michigan. Additional work may be done with markers that target the Rychc gene for PVY resistance if the pedigrees of parental germplasm show Solanum chacoense background. The Michigan breeding program has 7 different advanced breeding lines that are being used as parents, USDA-ARS, Aberdeen has approximately 40 resistant lines and the Maine breeding program typically uses 5-10 resistant clones as parents each year. True potato seed of the crosses will be shared between the programs and progeny will be grown out at multiple sites. The progeny will be selected for heritable tuber traits in the first field year, then DNA will be extracted from tubers or leaves of the year 1 selections for PCR-based PVY screening. Those selections that are positive for the PVY marker will be further advanced in the breeding programs for more intensive selection and evaluation.
